For me it is not so much whether or not there is a supreme being which makes life meaningful or not. The individual makes his or her life meaningful or not. The opportunities to create yourself as the human being you believe you should be are there. First you need to think long and hard on the kind of human being you should be. That has far less to do with money and status than it does with your character in my opinion. It certainly can be a challenging process though. In the world of relativity, the physical world, both good and evil exists. That is by design. Good cannot exist without it's opposite, "Evil". Courage does not exist without Fear. Honesty does not exist without it's opposite, Lies. In the spiritual or soul realm life is much easier but not without it's own challenges as I understand it.
Here the soul seeks to to put the physical you in a position where you choose the best, most honorable path for your progression. Sometimes that decision and continuing to make that decision consistently is quite difficult. But we need to forgive ourselves and forgive others too.
I believe in God but not as a supreme being separate from us or the Universe. I believe all of us are a part of God. I believe we are all souls with a spark of the divine presently housed in bodies. Yet our soul selves or higher selves, unfiltered by our brains and physical body limitations, is our true selves. We are here to experience a physical world of relativity, not available in the realm of pure souls. There is only light in the soul realm. No need for darkness there. No need for evil to overcome and choose against.
For me this life would lose "Most" of it's meaning if this life was all there was to our true existence in the Universe. It would certainly be less meaningful. I am convinced our soul, our higher self, leaves our dead body and continues on when we pass here on earth.
I am convinced there is no Hell. Hell was created by religions to scare the masses to keep them under control by the rich and powerful. The rich and powerful like the status quo, yet understand the masses could take both their money and power if the masses banded together to do so. Scare the Hell out of them with religion, tell them they are sinners (from birth) and need to be Good, Good, Good, while here or they will burn for all eternity in Hell. Part of being good is not killing the greedy rich and redistributing the wealth.
There is certainly an intelligent design to the Universe. Just look at nature and the miracle of our self healing bodies. I don't believe it was designed by a father figure of whom we were made in his physical image. That's almost as crazy as the Noah' Ark story. (Two of every beast on the boat? I don't think so.) Still there is most definitely an intelligent design to the universe. How was that set in motion? Or has it always been? It's all fascinating to me.
So life has more meaning if we continue on in an afterlife not if, or if not, there is a single supreme being running everything from beyond. That is my take on it. Yet life has the meaning you give it even if you don't believe in an afterlife.
